The expression levels of miRNA were determined by Real-time PCR and Northern blot . Those of protein were by Western blot analysis. Luciferase activity assay was used to verify the target genes of miRNAs. MTT assay was used to test the drug-resistant phenotype changes in cancer cells via overregulation of miRNAs. miRNA target prediction by computer algorithms . Soft agar assays were performed to determine the in vitro tumorigenicity of the cells. In vitro invasion assays were performed in BD BioCoat Matrigel chambers . Tumorigenicity assays in nude mice was used to test the role of miR-199a-5p .
